Effect of manganese toxicity on the indoleacetic Acid oxidase system of cotton.

چکیده
The effect of substrate manganese on tissue manganese levels and activity of the indoleacetic acid (IAA)-oxidase system of cotton (Gossypium hirsutum, L.) was investigated. A sand culture technique was used with 1, 3, 9, 27 and 81 mg manganese (MnSO(4)) per liter nutrient solution applied in various experiments. The following relationships held for both long-term (126 days) and short-term (12-14 days) exposures to manganese treatment: A) There was a direct relationship between substrate and tissue manganese. B) Only the 81 mg/liter Mn plants exhibited severe manganese toxicity symptoms. C) At the toxic level of manganese an increased IAA-oxidase activity and decreased IAA-oxidase inhibitor activity was observed. There was a direct relationship between degree of enzyme response and severity of visible symptoms. D) With the manganese toxicity plants, but none of the other treatments, extracts of the young leaves contained as much IAA-oxidase activity as extracts of much older leaves. E) Crude extracts from the plants grown with 81 mg manganese per liter solution, in contrast to those of other treatments, destroyed IAA without addition of MnCl(2) to the assay medium.A hypothesis is advanced stating that manganese toxicity symptoms in cotton are expressions of auxin deficiency caused by IAA-oxidase activity increased by the abnormal tissue levels of manganese.

Distribution of Indoleacetic Acid Oxidase and Inhibitors in Light-Grown Cotton.
Extracts of green plant tissues exhibiting IAAoxidase activity characteristically contain inhibitors of the enzyme (4, 11). The high concentration of such inhibitors prompted the discoverers of IAAoxidase to suggest that the system does not function in green tissue (16). Later, Galston and Dalberg (6) found that in etiolated tissue the in vitro IAAoxidase activity increases with age or IAA trea...
متن کاملStimulation of indoleacetic Acid oxidase and inhibition of catalase in cotton extracts by plant acids.
Activity of indoleacetic acid oxidase in partially purified extracts from cotton is stimulated by small amounts of malate, succinate, fumarate, and other plant acids. The stimulation is apparently due to inhibition of catalase, which is detectable in certain preparations. The lag phase of indoleacetic acid oxidation by crude preparations is eliminated by steps in processing which conceivably ei...
متن کاملThe effect of water stress on indoleacetic Acid oxidase in pea plants.
Activity of indoleacetic acid oxidase was shown to increase following a period of water stress. Two fractions of indoleacetic acid oxidase were extracted from plant extracts. Similarly, two protein peaks (determined by ultraviolet absorption) were isolated. One peak, associated with an indoleacetic acid oxidase peak, increased following water stress. The second peak, not associated with extract...

The enzymatic oxidlation of 3-indoleacetic aci(l (IAA) which was first described in detail by Tang and Bonner (16, 17), has been observed in several types of plants, e.g., fungi (13), ferns (14), monocotyledonous plants (20), and dicotyledonous plants (16, 17, 19). Although differences have been observed in regard to the enzyme's requirement for a phenolic cofactor, and in the pH optimum, the r...
متن کاملEffect of indoleacetic Acid and hydroxyproline on isoenzymes of peroxidase in wheat coleoptiles.
Indoleacetic acid at 0.017 millimolar inhibited the formation of three peroxidase isoenzymes in both soluble and wall-bound enzyme fractions of wheat coleoptile (Triticum vulgare) tissue. Hydroxyproline at 1 millimolar prevented the indoleacetic acid-induced inhibition. Indoleacetic acid oxidase activity in the soluble fraction was decreased by indoleacetic acid and was restored by hydroxyproli...
